چکیده

REFERENCES METHODS D-264 treatment partially rescued the loss of dopaminergic neurons in SN, inhibited the activation of microglia, and improve the impairment of behaviour caused by MPTP and lactacystin in pole and rotarod tests (data not shown here). The neuroprotective effects of D-264 shown in the present study may partly due to the upregulation of both BDNF and GDNF and the restoration of proteasomal activity . As a drug used in the clinic for other indications, we proposed that further studies on D-264 should be conducted in order to consider it as a novel therapy for PD. LEGENDS

Preclinical Study of Ibuprofen Loaded Transnasal Mucoadhesive Microemulsion for Neuroprotective Effect in MPTP Mice Model

Ibuprofen, a non-steroidal anti-inflammatory drug (NSAID), showed very promisingneuroprotection action, but it suffers from high first pass metabolism and limited abilityto cross blood brain barrier. Severe gastric toxicity following oral administration furtherlimits its utility. Hence, the aim of this study was to investigate whether ibuprofen loadedmucoadhesive microemulsion (MMEI) could enha...

Investigation of the Effect of Cycloserine on Motor Function in a Rat Model of Parkinson’s disease

Background: Previous studies have shown cycloserine to be neuroprotective in some neurodegenerative disorders. Objectives: To investigate the effect of cycloserine on motor function in Parkinson’s disease in a rat model. Materials and Methods: Fifty-six healthy male wistar rats were used in this study and were divided into seven groups according to receiving saline, low dose (i.e. 100 m...

Neuroprotective effects of lixisenatide and liraglutide in the MPTP mouse model of Parkinson’s disease

Glucagon-like peptide 1 (GLP-1) is a growth factor. GLP-1 mimetics are on the market as treatments for type 2 diabetes and are well tolerated. These drugs have shown neuroprotective properties in animal models of neurodegenerative disorders. In addition, the GLP-1 mimetic exendin-4 has shown protective effects in animal models of Parkinson’s disease (PD), and a clinical trial in PD patients sho...

Structural Modifications of Neuroprotective Anti-Parkinsonian (−)-N6-(2-(4-(Biphenyl-4-yl)piperazin-1-yl)-ethyl)-N6-propyl-4,5,6,7-tetrahydrobenzo[d]thiazole-2,6-diamine (D-264): An Effort toward the Improvement of in Vivo Efficacy of the Parent Molecule

In our overall goal to develop multifunctional dopamine D2/D3 agonist drugs for the treatment of Parkinson's disease (PD), we previously synthesized potent D3 preferring agonist D-264 (1a), which exhibited neuroprotective properties in two animal models of PD. To enhance the in vivo efficacy of 1a, a structure-activity relationship study was carried out. Competitive binding and [(35)S]GTPγS fun...
